数据非依赖性的扫描模式(Data-independent acquisition, DIA)是近几年来发展的一种新的质谱数据采集方式。可以对特定质量范围内的所有母离子进行碎裂,采集所有母离子的碎片离子信息进行蛋白定性和定量。目前流行的蛋白质组学研究手段,如iTRAQ\TMT、 Label-free、SILAC采用的都是数据依赖性的扫描模式(Data-dependent acquisition, DDA)。相对于DDA, DIA 具有更好的准确性和可重复性,是蛋白质组学研究的一大重要趋势。下图为 DIA 定量技术原理图:
